This is a working overview of 稳定性, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2026-03-31. Anything still debated is marked as such rather than presented as settled.
Reported outcomes in large trials include dose-dependent weight reduction and improvements in glycemic markers over periods ranging from several months to more than a year. Whether the compound alters long-term cardiovascular or renal outcomes is being examined in dedicated outcome studies, so those questions remain open. Labeling describes gastrointestinal effects such as nausea and diarrhea, which tend to appear during dose escalation. Discontinuation rates and the durability of effects after treatment stops vary across study populations and are still debated.
Tirzepatide is a synthetic peptide developed as a dual agonist at the glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 receptors. Its structure is built on a GIP-derived backbone with non-natural amino acid substitutions and a fatty diacid side chain that promotes albumin binding and slows clearance. That modification supports once-weekly subcutaneous dosing. Registrational trial programs reported reductions in body weight and glycated hemoglobin alongside the drug's glycemic effects.
Both receptors are class B G protein-coupled receptors that signal largely through Gs-mediated cyclic AMP production. Activation within pancreatic islets increases glucose-dependent insulin secretion and suppresses glucagon release when glucose is elevated. Outside the pancreas, signaling in the central nervous system and gut appears to influence appetite and gastric emptying. The relative contribution of each receptor to observed clinical effects remains under investigation, and the two pathways are not simply additive in practice.
Lyophilized material is generally held at -20 degrees Celsius or lower, desiccated and protected from light, where it remains stable for extended periods. Reconstituted or ready-to-use solution is usually kept at 2 to 8 degrees Celsius with minimal agitation. Repeated freeze-thaw cycles should be avoided because they promote aggregation and reduce the soluble monomer fraction. Shipment of frozen solid commonly uses dry ice, while refrigerated liquid moves with validated cold packs. Stability beyond documented periods is not established.
Characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity and related-substance profiling, with ultraviolet detection near 214 nanometers. Mass spectrometry confirms molecular mass and reveals modifications such as oxidation or deamidation. Peptide mapping after enzymatic digestion verifies the amino acid sequence, while amino acid analysis supplies compositional data. Circular dichroism and infrared spectroscopy are used to assess secondary structure, particularly the alpha-helical content that influences aggregation behavior in solution.
| Property | Value | Notes |
|---|---|---|
| Molecular class | Synthetic 39-residue peptide | GIP-derived backbone with non-natural residues |
| Molar mass | Approximately 4.8 kDa | Peptide chain plus linker and lipid modifications |
| Appearance | White to off-white powder | Typical of lyophilized research material |
| Solubility | Soluble in water and aqueous buffer | Practically insoluble in nonpolar solvents |
| Storage temperature | Typically −20 °C for dry powder | Solutions usually held at 2–8 °C short term |
Purified material is typically handled as a lyophilized powder kept at or below minus twenty degrees Celsius, shielded from light and moisture. In that state the solid remains stable for extended periods, although repeated freeze-thaw cycling can encourage aggregation. Once dissolved, aqueous solutions are less durable and are generally held cold and used within a brief window. Buffer composition, pH and ionic strength all influence degradation rates, and mildly acidic to neutral conditions are commonly examined. Actual shelf life depends on formulation, concentration and container, so stability limits are established experimentally rather than assumed.
Verification of research-grade material involves checking purity, sequence and counter-ion content against a certificate of analysis. Reported purity figures usually reflect chromatographic area percentage and do not by themselves establish biological activity. Independent laboratories may repeat mass confirmation and peptide mapping to detect substitutions or truncations. Open questions concern how residual solvents, trace metals and subtle conformational variants affect measured behavior, and how consistently different suppliers define their specifications. Documentation of analytical methods matters as much as the headline purity number when results are compared across studies.
Routine characterization relies on reversed-phase high-performance liquid chromatography, often coupled to mass spectrometry, to confirm identity and estimate purity. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates appended groups such as the fatty acid chain. Size-exclusion chromatography detects aggregates and fragments, while ion-exchange chromatography resolves charge variants. Circular dichroism and nuclear magnetic resonance supply secondary and higher-order structural information in research settings. No single technique covers every attribute, so laboratories combine orthogonal methods and compare outcomes against a reference standard where one exists.
CCK also causes the increased production of hepatic bile, and stimulates the contraction of the gall bladder and the relaxation of the sphincter of Oddi (Glisson's sphincter), resulting in the delivery of bile into the duodenal part of the small intestine. Bile salts form amphipathic lipids, micelles that emulsify fats, aiding in their digestion and absorption.

Liberation – the process of active pharmaceutical ingredients (API) separating from its pharmaceutical formulation. See also IVIVC. Absorption – the process of a drug entering into systemic circulation from the site of administration. Distribution – the dispersion or dissemination of substances throughout the fluids and tissues of the body. Metabolism (or biotransformation, or inactivation) – the chemical reactions of the drug and irreversible breakdown into metabolites (e.g. by metabolic enzymes such as cytochrome P450 or glucuronosyltransferase enzymes). Excretion – the removal of the substance or metabolites from the body. In rare cases, some drugs irreversibly accumulate in body tissue. Some textbooks combine the first two phases as the drug is often administered in an active form, which means that there is no liberation phase. Others include a phase that combines distribution, metabolism and excretion into a disposition phase. Other authors include the drug's toxicological aspect in what is known as ADME-Tox or ADMET. The two phases of metabolism and excretion can be grouped together under the title elimination. The study of these distinct phases involves the use and manipulation of basic concepts in order to understand the process dynamics.
The mean improvement in scores with dextromethorphan/bupropion was statistically significant but not clinically significant relative to placebo at all assessed timepoints including at the end of week 1, although at the end of the study some patients did have clinically significant improvement. In Study 2 (STRIDE-1), dextromethorphan/bupropion was compared with bupropion alone in another randomized controlled trial. The dose of bupropion in the study was lower than the target dose recommended for clinical practice. In this study, dextromethorphan/bupropion showed significantly greater improvement than bupropion alone in the first two weeks of treatment but not by week 6 of treatment in people with major depressive disorder. The baseline scores were 33.4 points with dextromethorphan/placebo and 33.2 points with placebo, while the score reductions at week 1 were 5.2 points on the MADRS with dextromethorphan/bupropion and 3.6 points with bupropion (a 1.6-point difference), at week 2 were 8.0 points with dextromethorphan/bupropion and 6.1 points with bupropion (a 1.9-point difference), and at week 6 were 11.6 points with dextromethorphan/bupropion and 9.4 points with bupropion (a 2.2-point difference). On the basis of this trial, the FDA concluded that dextromethorphan contributes to the apparent antidepressant effects of dextromethorphan/bupropion.
6 December Scientists provide a mechanistic biological explanation for why upper respiratory tract infections are more prevalent in the cold or winter season, previously largely explained only by behavioral and environmental variations. They found cold exposure impairs extracellular vesicle (EV) swarm–mediated antiviral immunity in the nose – fewer EVs are secreted and they become less effective. Impossible Metals announces its first underwater robotic vehicle, 'Eureka 1', has completed its first trial of selectively harvesting polymetallic nodule rocks from the seabed nearly without harming the environment (as with other seabed mining) to help address the rising global need for metals for renewable energy system components, mainly batteries. Researchers propose "significantly increasing freshwater through the capture of humid air over oceans" to address present and, especially, future water scarcity/insecurity. Emulate researchers assess advantages of using liver-chips predicting drug-induced liver injury which could reduce the high costs and time needed in drug development workflows/pipelines, sometimes described as the pharmaceutical industry's "productivity crisis". 7 December Scientists report that two-million years old genetic material was found in Greenland, and is currently considered the oldest DNA discovered so far. Scientists propose a new supergroup of eukaryotes, termed Provora – predators of other microorganisms which have been overlooked due to numerical rarity.

=== Pharmacokinetics === LC–MS is widely used in the field of bioanalysis and is specially involved in pharmacokinetic studies of pharmaceuticals. Pharmacokinetic studies are needed to determine how quickly a drug will be cleared from the body organs and the hepatic (liver) blood flow. MS analyzers are useful in these studies because of their shorter analysis time, and higher sensitivity and specificity compared to UV detectors commonly attached to HPLC systems. One major advantage is the use of tandem MS–MS, where the detector may be programmed to select certain ions to fragment. The measured quantity is the sum of molecule fragments chosen by the operator. As long as there are no interferences or ion suppression in LC–MS, the LC separation can be done quite swiftly.
Mimoun "Rocky" Kaabouni, who was nicknamed "The Moroccan Escobar", was born in Ben Taïeb, a village in the province of Nador. He grew up in Hoensbroek, in the Netherlands, where he and his brother owned a coffeeshop named Happy Days. While often visiting Amsterdam, he eventually started meeting big names within the Moroccan mafia with whom he maintained regular contact. Mimoun left the Netherlands for southern Spain and Romania, where he founded his own criminal organisation made up of Lithuanians and Colombians. Mimoun, who was already bringing in tons of cocaine through the ports of Antwerp, Rotterdam, Amsterdam, and Algeciras, wanted to create a new route in Romania through the port of Constanța. He is currently serving a 19-year prison sentence in Romania for a record seizure of 2.5 tons of cocaine (the biggest seizure of cocaine in the country's history). Mimoun was arrested in 2016 when he tried to leave Spain through the Málaga Airport after the seizure in Romania, which was worth €600 million. It was one of the largest drug seizures in Eastern European history.
== Synthesis == Endorphin precursors are primarily produced in the pituitary gland. All three types of endorphins are fragments of the precursor protein proopiomelanocortin (POMC). At the trans-Golgi network, POMC binds to a membrane-bound protein, carboxypeptidase E (CPE). CPE facilitates POMC transport into immature budding vesicles. In mammals, proprotein convertase 1 (PC1) cleaves POMC into adrenocorticotropin (ACTH) and beta-lipotropin (β-LPH). β-LPH, a pituitary hormone with little opioid activity, is then continually fragmented into different peptides, including α-endorphin, β-endorphin, and γ-endorphin. Proprotein convertase 2 (PC2) is responsible for cleaving β-LPH into β-endorphin and γ-lipotropin. Formation of α-endorphin and γ-endorphin results from proteolytic cleavage of β-endorphin.
Similar to propranolol with an extra contraindication for hyperthyroidism. In patients with thyrotoxicosis, possible deleterious effects from long-term use of pindolol have not been adequately appraised. Beta-blockade may mask the clinical signs of continuing hyperthyroidism or complications, and give a false impression of improvement. Therefore, abrupt withdrawal of pindolol may be followed by an exacerbation of the symptoms of hyperthyroidism, including thyroid storm. Pindolol has intrinsic sympathomimetic activity and is therefore used with caution in angina pectoris.

== Future research == Many aspects of food noise remain to be studied. Multiple future directions for research have been proposed, including the extent to which, how, and why GLP-1 RA drugs quiet food noise, how individuals’ experience of food noise relates to cognitive and physiological processes, demographic characteristics of those most susceptible to food noise, possible additional therapeutic approaches for managing food noise, and how public health policies may be implemented alongside food packaging, display, and information requirements to reduce the incidence of food noise.
== Mobile phases == A mobile phase in RP-LC consists of mixtures of water or aqueous buffers, to which further solvents are added, to elute analytes from a reversed-phase column in a selective manner. The added solvents must be miscible with water. These are also called "modifiers", since they modify the polarity of the mobile phase. Water is the most polar solvent in the reversed phase mobile phase. To increase the elution strength of the mobile phase, one usually add modifiers to lower its polarity. Common modifiers include acetonitrile (ACN), methanol (MeOH), 2-propanol (isopropanol, IPA), ethanol (EtOH) and tetrahydrofuran (THF).
=== Development of synthetic AAS === The development of muscle-building properties of testosterone was pursued in the 1940s, in the Soviet Union and in Eastern Bloc countries such as East Germany, where steroid programs were used to enhance the performance of Olympic and other amateur weight lifters. In response to the success of Russian weightlifters, the U.S. Olympic Team physician John Ziegler worked with synthetic chemists to develop an AAS with reduced androgenic effects. Ziegler's work resulted in the production of metandienone, which Ciba Pharmaceuticals marketed as Dianabol. The new steroid was approved for use in the U.S. by the Food and Drug Administration (FDA) in 1958. It was most commonly administered to burn victims and the elderly. The drug's off-label users were mostly bodybuilders and weight lifters. Although Ziegler prescribed only small doses to athletes, he soon discovered that those having used metandienone developed enlarged prostates and atrophied testes. AAS were placed on the list of banned substances of the International Olympic Committee (IOC) in 1976, and a decade later, the committee introduced "out-of-competition" doping tests because many athletes used AAS in their training period rather than during competition.

It acts as an agonist at both the GIP and GLP-1 receptors, two related class B G protein-coupled receptors. This dual activity distinguishes it from single-receptor GLP-1 agonists. The clinical consequences of engaging both receptors are still being characterized.
It is given by subcutaneous injection, generally on a weekly schedule. Dosing usually begins low and increases in steps to limit gastrointestinal side effects. Formulated product is supplied as a ready-to-use pen or vial in most markets.
No. Receptor engagement is documented, but how central appetite circuits, gut signals, and insulinotropic effects combine is incompletely resolved. Investigators continue to separate GIP-driven from GLP-1-driven contributions in animal and human models.
Reversed-phase high-performance liquid chromatography is the standard approach, separating the main peak from related impurities. Ultraviolet detection near 214 nanometers captures the peptide backbone. Mass spectrometry is then used alongside chromatography to confirm identity and detect covalent modifications.
