=== Level design === Lost Coast was conceived as a part of the Highway 17 chapter in Half-Life 2 (Highway 17's development name was "Coast", hence the name "Lost Coast"), but was discarded during development. It features storyline details that were removed from Half-Life 2, such as the headcrab artillery launchers. Each area was designed with a specific purpose. An Eastern Orthodox architectural style was chosen for the monastery, as buildings of this type "are very colorful and have a large variety of materials" and are "often lit naturally, with extremes of darkness and brightness", providing an ideal showcase for the HDR lighting effects. Valve also thought a monastery would provide a starker contrast between old human architecture and futuristic Combine technology inside it. The cliffside that leads to the monastery had a gameplay-oriented purpose, and was meant to emulate a similar cliffside combat scene in the original Half-Life (1998). The cliffside also forces the player to be observant of threats from above and below, breaking from normal horizontal combat. The monastery's courtyard was designed as an area where the player recovers from the cliffside combat, while also presenting a contained combat arena later in the level in which the player must hold their ground while they are attacked from multiple directions.
=== Genotypic identifications === For bacterial identifications, microbiologists sequence the 16S rRNA gene and for fungal identifications, sequence the ITS regions. Both regions are part of the ribosomal operon so they are well-conserved but provide enough variation to allow for speciation. Accurate identifications require high quality sequence data, a robust data analysis, and a broad microbial database of known organisms. It is also useful to use a Neighbor Joining tree or some other phylogenetic approach to make the identification.

It occurs naturally mainly in the minerals celestine and strontianite, and is mostly mined from these. == History == Mast cells were first described by Friedrich von Recklinghausen. In 1863 he reported the presence of granulated cells in connective tissues, observed in unstained cells of various species. They were later rediscovered and named in 1878 when Paul Ehrlich described the cells in terms of their unique staining characteristics and large granules. These granules led him to the incorrect belief that they existed to nourish surrounding tissue, so he named them Mastzellen or well-fed cells (from German Mast 'fattening'). In 1937, Holmgren and Wilander found that tissues rich in mast cells also contained large amounts of heparin. In 1952, Riley and West identified mast cells as a storage location for histamine. That mast cells released both heparin and histamine was demonstrated by Rocha e Silva in 1947. As a result of such work MCs became a major focus of allergy research. By 1999, mast cells were considered to be critical sentinel cells in the immune system.

== Chemical synthesis == A variety of synthetic approaches have progressively improved the efficiency and stereoselectivity of protolichesterinic acid synthesis while developing new methodologies for constructing similar lactone-containing natural products. The first total synthesis of dl-protolichesterinic acid was reported in 1958 by Eugene van Tamelen and Shirley Bach. Their synthesis involved a four-step route: conversion of methyl 2-hexadecenoate to methyl 3-tridecylglycidate, ring-opening with dimethyl malonate anion, formation of the lactonic diacid salt, and α-methylenation using formaldehyde and diethylamine. The synthetic material matched natural protolichesterinic acid by infrared spectroscopy and chemical transformations. In 1993, Murta, de Azevedo and Greene achieved the first synthesis of (-)-protolichesterinic acid, establishing its absolute stereochemistry as (2S,3R). Their approach employed a facially selective 2+2 cycloaddition of dichloroketene with an enantiopure O-alkyl enol ether as the key step, completing the synthesis in 11 steps with 17% overall yield. Mandal, Maiti and Roy reported a stereoselective synthesis in 1998 using radical cyclization of epoxides. ==== Urquía Carreño sends restitution payments ==== In August, 2025, government agents from the Ministry of Justice showed up at the National Masonic Temple with some bankers, and delivered 1 million Cuban pesos to the Grand Lodge. They informed the Grand Lodge that this money had been discovered during the course of the police investigation into Urquía Carreño, and that they should expect 3 million more pesos soon after. Filema Duarte and the office of the Grand Master published Special Circular No. 127, in which they asserted that the accountant Gertrudis Mena had digitally manipulated and falsified bank statements to steal and embezzle large sums of money, of which former Grand Master Urquía Carreño had been an accomplice. The total volume of embezzlement, not including the theft of the Llansó fund, was valued at around US$40,000. While many Freemasons praised the justice system for prosecuting against Urquía Carreño, they also stated that they believed it was just a tactic used by Filema Duarte as a method of distraction from his own scandal. In September 2025, a second payment was received by the Grand Lodge of Cuba from Former Grand Master Urquía Carreño and Former Grand Treasurer Airam Cervera Reigosa, totaling 4 million Cuban pesos. Grand Master Filema Duarte announced that the total sum of 5 million pesos was deposited into the account of the Grand Lodge of Cuba. Technically, both deposits were made under the name Airam Cervera, but it was taken implicitly that this was repayment for what both men owed the Grand Lodge.
