GIP receptor raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.

At the receptor level, tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. Both belong to the class B family of G protein-coupled receptors and signal largely through cyclic AMP accumulation. The compound binds the two receptors with differing affinity, and the pattern of signaling at each site is described in the literature as biased rather than simply proportional to occupancy. Tissues carrying these receptors include pancreatic islets, adipose tissue, the central nervous system, and the gastrointestinal tract. The relative weight of each receptor population in producing metabolic effects continues to be studied.
Published work supports the view that engaging two incretin receptors produces changes in glucose handling and body weight larger than those seen with single-receptor activation. Why that difference arises is not fully settled. Open questions include how much of the observed weight effect depends on central versus peripheral signaling, and whether the two receptors form interacting complexes. Most reported findings come from controlled trials and animal models, and translation between species is imperfect. Further research is expected to refine these points over time.
Structural work on the molecule centers on a C20 fatty diacid moiety attached through a linker to the peptide backbone. This side chain promotes reversible binding to serum albumin, which slows renal clearance and supports a prolonged action profile. The peptide backbone incorporates aminoisobutyric acid substitutions that limit recognition by digestive enzymes. Together these modifications produce a molecule that is stable enough for subcutaneous delivery but still dependent on careful manufacturing control. Analytical characterization of the active pharmaceutical ingredient typically follows the conventions used for other synthetic peptides.
Tirzepatide is a synthetic peptide composed of 39 amino acids. It acts as a dual agonist at two incretin receptors, the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. The molecule was designed by modifying the native sequence of glucose-dependent insulinotropic polypeptide to improve metabolic stability and extend its circulation time. Its structure includes several non-natural amino acid residues and a fatty acid side chain. These features distinguish it from earlier single-receptor incretin analogs studied in the same period.

An extended fatty diacid moiety promotes binding to serum albumin, which slows renal clearance and extends the circulating half-life to roughly five days. That property supports once-weekly administration and largely explains the dosing interval described in clinical reports. Published data come mainly from large randomised programmes that evaluated glycaemic control and body weight over periods of many months. Long-term outcomes beyond those trial windows, including what happens after treatment stops, remain an active area of investigation.
Tirzepatide is a synthetic peptide built from thirty-nine amino acids. Its sequence is derived from native glucose-dependent insulinotropic polypeptide, or GIP, with several non-natural residues and a fatty diacid side chain attached through a linker. The molecule behaves as a dual agonist at two incretin receptors, GIP and GLP-1, instead of targeting a single receptor. This dual engagement separates it from earlier single-receptor incretin compounds and underpins most of its reported pharmacological activity.
Development began in the 2010s, when researchers modified a GIP-based scaffold to add GLP-1 activity and then attached the fatty diacid to lengthen its half-life. Clinical evaluation proceeded through large phase 3 programmes in type 2 diabetes and in obesity, and regulators in the United States cleared the compound for type 2 diabetes in 2022 and for chronic weight management in 2023. Several cardiovascular and metabolic outcome studies are still reporting, so the picture of long-term benefit and risk is incomplete. Approvals in other regions followed on different timelines.
Tirzepatide is a synthetic peptide of 39 amino acids that carries a C20 fatty diacid side chain attached through a linker. Its molecular formula is C225H348N48O68, and its molecular weight is about 4813 daltons. The compound belongs to the incretin mimetic class and is administered by subcutaneous injection. The fatty acid chain promotes binding to serum albumin, which slows renal clearance and extends the circulation time of the molecule. It was identified during screening of sequences derived from glucose-dependent insulinotropic polypeptide.
Tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor, making it a dual agonist rather than a selective agent. Engagement of the GLP-1 receptor is linked to glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. The relative contribution of the GIP arm remains an active research question; proposed roles include improved insulin sensitivity and altered adipose tissue handling. Receptor occupancy studies suggest the molecule interacts with both targets at circulating concentrations achieved during therapy.
Dual agonism at the GIP and GLP-1 receptors underlies the observed pharmacology. Activation of GLP-1 receptors raises glucose-dependent insulin release, lowers glucagon secretion, slows gastric emptying and reduces appetite. GIP receptor activation contributes additional effects on adipose tissue and on energy balance, and the combined action on appetite appears larger than either pathway alone in animal models. Signalling bias and the relative contribution of each receptor arm to weight-related effects remain areas of active investigation.
Structure-activity work shows that fatty acid length, linker chemistry and the position of acylation all influence albumin affinity and receptor potency. Plasma protein binding exceeds 99 percent, which restricts distribution and slows renal clearance. Degradation proceeds largely through general proteolysis and fatty acid oxidation rather than cytochrome P450 metabolism, so exposure to common oxidative drug interactions is limited. Whether these clearance routes vary meaningfully between individuals is not fully established.
The molecule is a synthetic 39-amino-acid peptide whose backbone derives from the sequence of human glucose-dependent insulinotropic polypeptide, with several substitutions that raise metabolic stability and shift receptor preference. A C20 fatty diacid is attached through a short linker to a lysine side chain, a modification that increases binding to serum albumin. The reported monoisotopic mass is approximately 4813 Da. Near neutral pH the peptide carries a net negative charge, and the lipid tail makes the molecule markedly more hydrophobic than the unmodified parent sequence.
The balance would, at least theoretically, actually decrease in size with heating to compensate for the lengthening of the hairspring. Through careful adjustment of the placement of the balance screws (brass or gold screws placed in the rim of the balance), a watch could be adjusted to keep time the same at both hot (100 °F (38 °C)) and cold (32 °F (0 °C)) temperatures. Unfortunately, a watch so adjusted would run slow at temperatures between these two. The problem was completely solved through the use of special alloys for the balance and hairspring which were essentially immune to thermal expansion. Such an alloy is used in Hamilton's 992E and 992B. Isochronism was occasionally improved through the use of a stopworks, a system designed to only allow the mainspring to operate within its center (most consistent) range. The most common method of achieving isochronism is through the use of the Breguet overcoil, which places part of the outermost turn of the hairspring in a different plane from the rest of the spring. This allows the hairspring to "breathe" more evenly and symmetrically. Two types of overcoils are found - the gradual overcoil and the Z-Bend. The gradual overcoil is obtained by imposing two gradual twists to the hairspring, forming the rise to the second plane over half the circumference; and the Z-bend does this by imposing two kinks of complementary 45 degree angles, accomplishing a rise to the second plane in about three spring section heights. The second method is done for esthetic reasons and is much more difficult to perform.
The enthesis (plural entheses) is the connective tissue which attaches tendons or ligaments to a bone. There are two types of entheses: fibrous entheses and fibrocartilaginous entheses. In a fibrous enthesis, the collagenous tendon or ligament directly attaches to the bone. In a fibrocartilaginous enthesis, the interface presents a gradient that crosses four transition zones:
On-line mass spectrometry was develop to solve some of the limitations and problem that develop from off-line analysis, such as evaporation and chemical reactions of particles in the filters during long analysis time. On-line Mass spectrometry solves these problems through the collection and analysis of aerosol particles in real time. On-line instruments are very portable and allow for spatial variability to be examined. These portable instruments can be put on many different platforms such as boats, planes, and mobile platforms (e.g. car trailers). An example of this is in the picture at the beginning with the instrumentation attached to an aircraft. Like off-line, on-line mass spectrometry has many different type of instruments, which can be broken up into two types; instruments that measures the chemistry of the particle ensemble (bulk measurement) and those that measure the chemistry of individual particles (single-particle measurement). Thus based on analytical need different instrumentation is used in analysis of the aerosol particles.
The following table illustrates variability in reference ranges of serum prolactin between some commonly used assay methods (as of 2008), using a control group of healthy health care professionals (53 males, age 20–64 years, median 28 years; 97 females, age 19–59 years, median 29 years) in Essex, England: An example of the use of the above table is, if using the Centaur assay to estimate prolactin values in μg/L for females, the mean is 168 mIU/L (7.92 μg/L) and the reference range is 71–348 mIU/L (3.35–16.4 μg/L). Hyperprolactinaemia, or excess serum prolactin, is associated with hypoestrogenism, anovulatory infertility, oligomenorrhoea, amenorrhoea, unexpected lactation and loss of libido in women and erectile dysfunction and loss of libido in men. input from rivers sulfate reduction and sulfide re-oxidation on continental shelves and slopes burial of anhydrite and pyrite in the oceanic crust. The primary natural source of sulfur to the atmosphere is sea spray or windblown sulfur-rich dust, neither of which is long lived in the atmosphere. In recent times, the large annual input of sulfur from the burning of coal and other fossil fuels has added a substantial amount of SO2 which acts as an air pollutant. In the geologic past, igneous intrusions into coal measures have caused large scale burning of these measures, and consequential release of sulfur to the atmosphere. This has led to substantial disruption to the climate system, and is one of the proposed causes of the Permian–Triassic extinction event. Dimethylsulfide [(CH3)2S or DMS] is produced by the decomposition of dimethylsulfoniopropionate (DMSP) from dying phytoplankton cells in the ocean's photic zone, and is the major biogenic gas emitted from the sea, where it is responsible for the distinctive “smell of the sea” along coastlines. DMS is the largest natural source of sulfur gas, but still only has a residence time of about one day in the atmosphere and a majority of it is redeposited in the oceans rather than making it to land. However, it is a significant factor in the climate system, as it is involved in the formation of clouds.

== Bulk properties == Thorium is a moderately soft, paramagnetic, bright silvery radioactive actinide metal that can be bent or shaped. In the periodic table, it lies to the right of actinium, to the left of protactinium, and below cerium. Pure thorium is very ductile and, as normal for metals, can be cold-rolled, swaged, and drawn. At room temperature, thorium metal has a face-centred cubic crystal structure; it has two other forms, one at high temperature (over 1360 °C; body-centred cubic) and one at high pressure (around 100 GPa; body-centred tetragonal). Thorium metal has a bulk modulus (a measure of resistance to compression of a material) of 54 GPa, about the same as tin's (58.2 GPa). Aluminium's is 75.2 GPa; copper's is 137.8 GPa; and mild steel's is 160–169 GPa. Thorium is about as hard as soft steel, so when heated it can be rolled into sheets and pulled into wire. Thorium is nearly half as dense as uranium and plutonium and is harder than both. Thorium has a magnetic susceptibility of 0.412 × 4π × 10−9 m3⋅kg−1 at room temperature. This susceptibility is mostly temperature-independent, however impurities and dopants can affect this value. It becomes superconductive below 1.4 K. Thorium's melting point of 1750 °C is above both those of actinium (1227 °C) and protactinium (1568 °C).
== History == Lixisenatide is a GLP-1 receptor agonist that was created by Zealand Pharma A/S of Denmark; in 2003 Zealand licensed it to Sanofi which developed the drug. Lixisenatide was approved by the European Commission on 1 February 2013. Sanofi submitted an NDA in the US, which was accepted for review by the US FDA in February 2013 but after discussions with the FDA about the cardiovascular safety data included in the package (starting in 2008, the FDA had required stronger CV safety data for new anti-diabetes drugs, following the controversy around the risks of Avandia) Sanofi decided to withdraw the NDA and wait for the results of a Phase III study that was scheduled to be completed in 2015. Sanofi resubmitted the application which the FDA accepted in September 2015, by which time Sanofi had lost the lead in the field of anti-diabetic drugs to Novo Nordisk. Lixisenatide received FDA approval on 28 July 2016. In 2010, Sanofi extended a license agreement it had with Zealand for lixisenatide to allow Sanofi to combine it with insulin glargine, which was Sanofi's best selling drug at the time, with sales of around €3 billion in 2009. Sanofi planned to start the Phase III trial that year. Sanofi submitted the new drug application in December 2015 for the combination and spent a US$245M priority voucher to gain a faster review, to try to outrace Novo Nordisk’s Xultophy, a similar combination drug of Novo's insulin Tresiba and Novo's GLP-1 agonist Victoza.
The primary sequence of this protein in Arabidopsis is listed below. Residues involved in catalytic activity are bolded. Mutagenesis of Asp428, which is crucial carboxylate ligand to Mg(2+) in the "ThDP motif", leads to a decrease in the affinity of AHAS II for Mg(2+). While mutant D428N shows ThDP affinity close to that of the wild-type on saturation with Mg(2+), D428E has a decreased affinity for ThDP. These mutations also lead to dependence of the enzyme on K(+).
An immigration stamp is an inked impression in a passport or other travel document, typically made by a rubber stamp upon entering or exiting a territory. Depending on the jurisdiction, a stamp can serve different purposes. For example, in the United Kingdom, an immigration stamp in a passport includes the formal leave to enter granted to a person subject to entry control. In other countries, a stamp activates or acknowledges the continuing leave conferred in the passport bearer's entry clearance. Under the Schengen system, a foreign passport is stamped with a date stamp which does not indicate any duration of stay. This means the person is deemed to have permission to remain for either 3 months or the period shown on their visa, if specified otherwise. Member states of the European Union are not permitted to place a stamp in the passport of a person who is not subject to immigration control. Stamping is prohibited because it imposes a control to which the person is not subject. Passport stamps may occasionally take the form of sticker stamps, such as entry stamps from Japan. Depending on nationality, a visitor may not receive a stamp at all (unless specifically requested), such as an EU or EFTA citizen travelling to an EU or EFTA country, Albania, or North Macedonia. Most countries issue exit stamps in addition to entry stamps. A few countries issue only entry stamps, including Canada, El Salvador, Ireland, Mexico, New Zealand, Singapore, United Kingdom and the United States of America.
polymerase Any of a class of enzymes which catalyze the synthesis of polymeric molecules, especially nucleic acid polymers, typically by encouraging the base pairing of free nucleotides with those of an existing complementary template strand. DNA polymerases and RNA polymerases are essential for DNA replication and transcription, respectively.

Rush added former Chicago keyboardist Loren Gold as a touring musician in late February 2026; his arrival to the band marked the first time since second guitarist Mitch Bossi's departure from Rush in 1972 that they have performed together as a quartet rather than a trio. In January 2026, Lee revealed in an interview with Music Radar that there was a possibility of new music from Rush upon completion of the tour, saying, "Who knows what'll happen, but I suspect some music will eventually come out", and added, "It would be fun to see what [Nilles] can do in a creative situation. Like, that would be fun. But it's all speculation until it isn't." When asked in August if the new line-up of Rush had already thought of recording new music, Lee said, "No. We have enough to do. We're still trying to remember the old songs and play them." On March 29, 2026, Rush gave a surprise performance at the 2026 Juno Awards, performing "Finding My Way" with their new line-up.

The study of human physiology began with Hippocrates in Ancient Greece, around 420 BCE, and with Aristotle (384–322 BCE) who applied critical thinking and emphasis on the relationship between structure and function. Galen (c. 129 – c. 216) was the first to use experiments to probe the body's functions. The term physiology was introduced by the French physician Jean Fernel (1497–1558). In the 17th century, William Harvey (1578–1657) described the circulatory system, pioneering the combination of close observation with careful experiment. In the 19th century, physiological knowledge began to accumulate at a rapid rate with the cell theory of Matthias Schleiden and Theodor Schwann in 1838, that organisms are made up of cells. Claude Bernard (1813–1878) created the concept of the milieu interieur (internal environment), which Walter Cannon (1871–1945) later said was regulated to a steady state in homeostasis. In the 20th century, the physiologists Knut Schmidt-Nielsen and George Bartholomew extended their studies to comparative physiology and ecophysiology. Most recently, evolutionary physiology has become a distinct subdiscipline.
== Vasoactive Intestinal Peptide (VIP) and SARS-CoV-2 == VIP is highly localised in lungs and binds with alveolar type II (AT II) cells via VPAC1 receptor. AT II cells constitute only 5% of pulmonary epithelium. Angiotensin Converting Enzyme 2 (ACE 2) surface receptors are present in AT II cells. AT II cells produces surfactant and plays an important role in the maintenance of type 1 epithelial cells. SARS-CoV-2 enters into AT II cells by binding to ACE 2 surface receptors with its spike protein.
Research on the viability of this new technique, as well as many others, as a means of monitoring the levels of rBST in dairy cattle continues to be an issue of practicality, profitability, and humaneness towards livestock in the dairy industry.

It is a synthetic peptide and a dual agonist of two incretin receptors. It is not a small molecule, and it is not structurally related to the older single-receptor peptide agonists.
The C20 fatty diacid promotes tight binding to serum albumin. That binding reduces renal clearance and extends circulation time compared with an unmodified peptide of similar length.
It is not fully established. Studies indicate that both receptors contribute to the observed effects, but the exact split between the two signaling pathways in humans remains an open question.
It activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. This dual activity separates it from agents that act on only one of the two receptors. The relative contribution of each receptor to clinical effects remains an open area of study.
