The peptide shares degradation routes common to modified peptides: deamidation of asparagine and glutamine residues, oxidation of methionine, and backbone hydrolysis under extreme pH. Lyophilized material is generally more stable than a solution, and residual water content directly affects the rate of hydrolysis. In liquid form, aggregation and visible particles can appear after agitation or repeated freeze-thaw cycles. Stability studies therefore track monomer content, aggregate content, and potency over months under defined temperature and humidity.
Cold-chain handling is standard for formulated product, with dry powder stored frozen and ready-to-use solutions refrigerated. Light exposure is minimized because photodegradation of certain amino acid side chains is possible. Shipping and temperature-excursion studies are used to establish whether short deviations affect quality attributes. Documentation supplied with research material usually includes a certificate of analysis listing purity, identity confirmation, and water or residual solvent content. Users are expected to confirm that material meets the stated specification before use.
Regulatory and quality discussions place the peptide within established guidance for synthetic peptides and biologics. Forced degradation studies, in which samples are exposed to heat, acid, base, peroxide, and light, identify likely degradation products and validate the selectivity of analytical methods. Reference standards allow comparison across laboratories and production batches. Purity specifications reported in the literature usually combine chromatographic purity with mass confirmation. Which impurity thresholds are meaningful for long-term behavior is still debated, and no single universal specification has been adopted across all jurisdictions.
Routine characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity assessment, usually with ultraviolet detection near 214 nanometers. Intact mass measurement by liquid chromatography coupled to mass spectrometry confirms molecular identity against a theoretical value. Sequence-level confirmation uses enzymatic digestion followed by tandem mass spectrometry, an approach known as peptide mapping. Amino acid analysis gives an independent check on composition. Circular dichroism spectra are used to estimate helical content in aqueous buffer.
Stability depends strongly on physical form. The dry powder is generally regarded as stable for extended periods when held at or below minus twenty degrees Celsius in a sealed, desiccated container. In solution, degradation pathways include deamidation of asparagine and glutamine residues, oxidation of methionine, and aggregation. Reaction rates for these pathways rise with temperature. Repeated freezing and thawing of solutions promotes aggregation, and light exposure can accelerate some oxidative changes. Buffer composition and pH influence which pathway dominates at a given temperature.

The GIP receptor is expressed in pancreatic islets, adipose tissue, and the central nervous system, while GLP-1 receptors are found in pancreatic islets, the gastrointestinal tract, and the brain. Activation of both receptors can enhance glucose-dependent insulin secretion and reduce glucagon release. The relative contribution of each receptor to the overall pharmacological effect remains an area of ongoing investigation. Preclinical studies suggest that GIP receptor agonism may modulate appetite and energy balance, but the precise mechanisms in humans are not fully established.
In clinical research, tirzepatide has been studied in randomized controlled trials for glycemic control and body weight reduction. These trials typically measure changes in hemoglobin A1c and body weight over periods of several months. The drug is administered by subcutaneous injection, and its pharmacokinetic profile supports once-weekly dosing. Post-marketing surveillance continues to evaluate long-term outcomes and rare adverse events.
Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
== Medical uses == Vildagliptin is indicated, as an adjunct to diet and exercise, to improve glycemic control in adults with type 2 diabetes as monotherapy in people in whom metformin is inappropriate due to contraindications or intolerance; and in combination with other medicinal products for the treatment of diabetes, including insulin, when these do not provide adequate glycemic control.
In December 2008, Dmitry Medvedev said that Russian defense minister Anatoly Serdyukov informed him around 01:00 on 8 August 2008 that Georgia had declared war on South Ossetia. He declared that Russia was preparing for the war with Georgia. He said that in August 2008, "I did not hesitate even for a second and gave order" to begin military action against Georgia. The Georgian foreign ministry said that Medvedev's statement was a "plea of guilty". In August 2011, Dmitry Medvedev said: "The moment of truth for me, as I realized later while analyzing those events in hindsight over and over again, came with the visit by Secretary of State Condoleezza Rice." He accused Saakashvili of dropping all contacts with the Russian governments following Rice's visit because he was preparing to start the war. Medvedev also said that he talked to Prime Minister Vladimir Putin only the following day after he had made a decision to attack Georgia on his own.

In late January, several CDC websites, pages, and datasets related to HIV and STI prevention, and LGBT and youth health became unavailable for viewing. In mid-February, around 1,300 CDC employees were laid off. In April, it was reported that among the reductions were the elimination of the Freedom of Information Act team, the Division of Violence Prevention, labs involved in testing for antibiotic resistance, and the team responsible for recalls of hazardous infant products. Additional cuts affected the technology branch of the Center for Forecasting and Outbreak Analytics, which was established during the COVID-19 pandemic. On June 25, 2025, Kennedy announced that the U.S. was stopping its donations to the Gavi vaccine alliance, until Gavi can better demonstrate vaccine safety. The United States had been providing approximately 13 percent of Gavi's budget. On August 5, 2025, Secretary Kennedy announced that he was stopping 22 vaccine projects using mRNA technology, including Covid, RSV, and bird flu. A critic of this funding halt pointed out that mRNA vaccines have the potential for faster roll-out. Freezing point depression osmometers may also be used to determine the osmotic strength of a solution, as osmotically active compounds depress the freezing point of a solution. This is the most common method in clinical laboratories because it is the most accurate and simple method. Vapor pressure osmometers determine the concentration of osmotically active particles that reduce the vapor pressure of a solution. Membrane osmometers measure the osmotic pressure of a solution separated from pure solvent by a semipermeable membrane. Historically, the early osmometer (Pfeffer cell, 1877), belonged to the membrane type. Osmometers are useful for determining the total concentration of dissolved salts and sugars in blood or urine samples. Osmometry is also useful in determining the molecular weight of unknown compounds and polymers. Osmometry is the measurement of the osmotic strength of a substance. This is often used by chemists for the determination of average molecular weight. Osmometry is also useful for estimating the drought tolerance of plant leaves.
In a typical gas chromatography analysis, isotopic dilution can decrease the uncertainty of the measurement results from 5% to 1%. It can also be used in mass spectrometry (commonly referred to as isotopic dilution mass spectrometry or IDMS), in which the isotopic ratio can be determined with precision typically better than 0.25%.
Finally, Alexander Shulgin has suggested that nutmeg might be a serotonergic psychedelic via conversion of active constituents into psychedelic amphetamines along with toxic effects from other constituents, though this has not been supported. Nutmeg intoxication has also been confused with those of phencyclidine (PCP) and alcohol as well as likened in some respects to that of alcohol. A clinical study of an isolated fraction of nutmeg that appears to have contained a mixture of myristicin, elemicin, and perhaps a small amount of methylisoeugenol was conducted and published in 1961. A dose of 400 mg of the mixture (equivalent to about 40 g whole nutmeg) was given orally. It produced symptoms suggestive of psychoactive effects such as mild stimulation in 6 of 10 individuals and definite psychoactive effects in 4 of 10 individuals, but failed to exactly reproduce the intoxication characteristic of nutmeg. Effects included euphoria, restlessness, weakness, nervousness, tremor, anxiety, tachycardia, gastrointestinal discomfort, nausea, and others. In the same study, it was found that 10 grams nutmeg with the volatile compounds removed produced no psychoactive effects but did cause in some instances undesirable side effects. It has been suggested that there might be a synergy between myristicin and other nutmeg constituents, for instance other individual allylbenzenes. Alternatively, other constituents may be responsible for the psychoactive and hallucinogenic effects instead. Shulgin reported one other constituent, methoxyeugenol, to be inactive at doses of up to 10 mg orally.
Cosmetic procedures that have trended on TikTok include rhinoplasties, buccal fat removals, and botox injections. Cosmetic procedures sometimes trend in the form of an internet challenge on TikTok. The "#Nosejobcheck" challenge involves users posting videos of their noses before and after their rhinoplasties, with a specific background sound for the challenge used in the videos.

In polymer chemistry, a random coil is a conformation of polymers where the monomer subunits are oriented randomly while still being bonded to adjacent units. It is not one specific shape, but a statistical distribution of shapes for all the chains in a population of macromolecules. The conformation's name is derived from the idea that, in the absence of specific, stabilizing interactions, a polymer backbone will "sample" all possible conformations randomly. Many unbranched, linear homopolymers—either in solution, or above their melting temperatures— assume (approximate) random coils.
==== Law 116, Puerto Rico ==== In 1937, Law 116 legalized sterilization in Puerto Rico. This law implemented Eugenics Boards within 32 states that oversaw compulsory sterilizations. More specifically, the Puerto Rican Eugenics Boards reviewed and confirmed petitions from the government and private entities to inflict sterilizations amongst the perceived "insane", "feeble minded", "diseased", and "dependent". The purpose of the Puerto Rican Eugenics Board was to regulate the reproductive capacities of "socially inferior" and perceived undesirable Puerto Ricans. This led the Puerto Rican Eugenics Board to approve 97 sterilizations before it was dissolved. Additionally, a large purpose of Law 116 was to further the science of eugenics and incite economic growth. Law 116 was the result of an increase of curiosity and political support for the science of eugenics. It was legitimized by the belief that Puerto Rico was a failing economy that consisted of "unfit" people that should be addressed by decreasing the population density through the means of forced sterilizations. Therefore, population control programs became institutionalized as well as federally subsidized. Funds from both the U.S. government and private investors enabled the last eugenics sterilization law passed under United States territorial jurisdiction. It also legalized state-mandated and forced sterilizations, which further exploited Puerto Ricans.
=== Diet === Research by Alan Ebringer at King's College in London, beginning in the 1980s, implicates overgrowth of the bacterium Klebsiella pneumoniae in the symptoms of ankylosing spondylitis. The body produces antibodies that attack Klebsiella pneumoniae. Enzymes made by the bacterium resemble human proteins, including three types of collagen (I, III, IV) and the HLA-B27 complex of glycoproteins. The antibodies therefore attack these human proteins, producing the symptoms of ankylosing spondylitis. Ebringer and others recommend low-starch or no-starch diets.

Reversed-phase liquid chromatography with ultraviolet detection is the usual approach, frequently combined with mass spectrometry for identity. Purity is reported as the area percentage of the main peak. Related impurities eluting near the main peak are usually summed and reported separately.
Removing water slows hydrolysis and limits aggregation, so dry powder retains its quality attributes longer than a solution. Suppliers define a shelf life and retest date for the dried form at specified temperatures. Once dissolved, the practical working lifetime shortens considerably.
It generally lists appearance, identity by mass, purity by chromatography, water or residual solvent content, and the methods used. Storage recommendations and a retest date are commonly included. Values are reported against a supplier specification rather than a single universal standard.
Peptide mapping with tandem mass spectrometry is the standard approach. The peptide is digested with an enzyme such as trypsin, and the resulting fragments are matched against the expected sequence.
