Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.
Characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity and related-substance profiling, with ultraviolet detection near 214 nanometers. Mass spectrometry confirms molecular mass and reveals modifications such as oxidation or deamidation. Peptide mapping after enzymatic digestion verifies the amino acid sequence, while amino acid analysis supplies compositional data. Circular dichroism and infrared spectroscopy are used to assess secondary structure, particularly the alpha-helical content that influences aggregation behavior in solution.
Common degradation routes include hydrolysis of labile amide bonds, deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation. Aggregates can form during freeze-thaw cycling, at elevated pH, or when peptide concentration is high. Each route produces characteristic chromatographic or mass shifts that are tracked during stability studies. Whether a given minor impurity alters biological activity is often an open question, and specification limits are typically set on identity and purity rather than on functional data for trace species.
Lyophilized material is generally held at -20 degrees Celsius or lower, desiccated and protected from light, where it remains stable for extended periods. Reconstituted or ready-to-use solution is usually kept at 2 to 8 degrees Celsius with minimal agitation. Repeated freeze-thaw cycles should be avoided because they promote aggregation and reduce the soluble monomer fraction. Shipment of frozen solid commonly uses dry ice, while refrigerated liquid moves with validated cold packs. Stability beyond documented periods is not established.

Tirzepatide is a synthetic peptide of 39 amino acids engineered from the native glucose-dependent insulinotropic polypeptide sequence. Its structure incorporates several non-natural residues and a C-terminal segment derived from glucagon-like peptide-1, together with a C20 fatty diacid moiety attached through a linker. The lipophilic side chain promotes binding to serum albumin, which slows renal clearance after administration. The compound is classified as a dual incretin receptor agonist and is supplied as a lyophilized powder for reconstitution or as a preformulated solution, depending on the presentation.
The peptide activates two G protein-coupled receptors, GIPR and GLP-1R. Binding triggers adenylyl cyclase activity and raises intracellular cyclic AMP in pancreatic beta cells, which potentiates insulin release when glucose is elevated. Signaling in the central nervous system is associated with reduced appetite and lower energy intake, while effects on gastric emptying and glucagon secretion are also reported. Because activity at both receptors is retained, the pharmacological profile is often described as incretin-based rather than selective for a single receptor.
The molecule is a synthetic 39-amino-acid peptide whose backbone derives from the sequence of human glucose-dependent insulinotropic polypeptide, with several substitutions that raise metabolic stability and shift receptor preference. A C20 fatty diacid is attached through a short linker to a lysine side chain, a modification that increases binding to serum albumin. The reported monoisotopic mass is approximately 4813 Da. Near neutral pH the peptide carries a net negative charge, and the lipid tail makes the molecule markedly more hydrophobic than the unmodified parent sequence.
Dual agonism at the GIP and GLP-1 receptors underlies the observed pharmacology. Activation of GLP-1 receptors raises glucose-dependent insulin release, lowers glucagon secretion, slows gastric emptying and reduces appetite. GIP receptor activation contributes additional effects on adipose tissue and on energy balance, and the combined action on appetite appears larger than either pathway alone in animal models. Signalling bias and the relative contribution of each receptor arm to weight-related effects remain areas of active investigation.

==== Notes ==== a See Freeze brand § Freeze branding as a painless alternative to mulesing. b See the subsection on human cryobranding in Freeze brand § Usage. c Irwin reports that keeping the iron stationary when branding a dolphin's dorsal fin, rather than rocking it as with livestock, produced a markedly clearer result. d Although adequate marks appeared on the elephant seal's skin at the time of cryobranding none of the marks produced lasted more than 1 year. e Down feathers grew white after cryobranding but did not persist in adult Mallards nor affect the outer contour feathers when they grew in. According to Greenwood, citing Foulks: "melanocytes continually arise from undifferentiated melanoblasts in the dermis during feather growth. Apparently the presence of whitetipped down resulted from the destruction of melanocytes associated with the tip of the developing feather, which were later replaced with active melanocytes before the deposition process terminated. Although birds were treated at ages of 5–10 days, and at various early stages of feather development, no apparent effect on pigmentation of contour feathers occurred." f Herpetologist Charles F. Smith described freeze branding as "[in]effective for bufonids [toads] and other taxa with granular skin surfaces." g All salamanders tested were seriously injured, and some later died. == Function == Glucagon generally elevates the concentration of glucose in the blood by promoting gluconeogenesis and glycogenolysis. Glucagon also decreases fatty acid synthesis in adipose tissue and the liver, as well as promoting lipolysis in these tissues, which causes them to release fatty acids into circulation where they can be catabolised to generate energy in tissues such as skeletal muscle when required. Glucose is stored in the liver in the form of the polysaccharide glycogen, which is a glucan (a polymer made up of glucose molecules). Liver cells (hepatocytes) have glucagon receptors. When glucagon binds to the glucagon receptors, the liver cells convert the glycogen into individual glucose molecules and release them into the bloodstream, in a process known as glycogenolysis. As these stores become depleted, glucagon then encourages the liver and kidney to synthesize additional glucose by gluconeogenesis. Glucagon turns off glycolysis in the liver, causing glycolytic intermediates to be shuttled to gluconeogenesis. Glucagon also regulates the rate of glucose production through lipolysis. Glucagon induces lipolysis in humans under conditions of insulin suppression (such as diabetes mellitus type 1). Glucagon production appears to be dependent on the central nervous system through pathways yet to be defined. In invertebrate animals, eyestalk removal has been reported to affect glucagon production. Excising the eyestalk in young crayfish produces glucagon-induced hyperglycemia.
=== Hormonal interactions === Progesterone has a number of physiological effects that are amplified in the presence of estrogens. Estrogens through estrogen receptors (ERs) induce or upregulate the expression of the PR. One example of this is in breast tissue, where estrogens allow progesterone to mediate lobuloalveolar development. Elevated levels of progesterone potently reduce the sodium-retaining activity of aldosterone, resulting in natriuresis and a reduction in extracellular fluid volume. Progesterone withdrawal, on the other hand, is associated with a temporary increase in sodium retention (reduced natriuresis, with an increase in extracellular fluid volume) due to the compensatory increase in aldosterone production, which combats the blockade of the mineralocorticoid receptor by the previously elevated level of progesterone.

Brec Bassinger as Courtney Whitmore / Stargirl:A high school student from Los Angeles who finds a powerful weapon, the Cosmic Staff, and becomes the superheroine Stargirl. As Stargirl, she also becomes the leader of the second incarnation of the Justice Society of America (JSA). In preparation for the role, Bassinger did not look at the character's previous portrayals by Britt Irvin on Smallville and Sarah Grey on Legends of Tomorrow because they were depicted as "older, more mature version[s]" of Stargirl. Bassinger added that the series follows the comic Stars and S.T.R.I.P.E. more "in that she's young, high school, naive", which she wanted to bring that into her version of Stargirl. Maizie Smith portrays a five-year old Courtney. Yvette Monreal as Yolanda Montez / Wildcat II:A once popular student at Blue Valley High until a scandal made her an outcast and a disgrace to her Catholic parents. A skilled boxer, she becomes one of Courtney's friends and a member of the new JSA as the new Wildcat. Anjelika Washington as Beth Chapel / Doctor Mid-Nite II: A social reject and nerd who becomes one of Courtney's friends and a member of the new JSA as the new Doctor Mid-Nite. ==== MVP runner-up and second player with multiple 40–30 seasons (1999) ==== Bagwell's former high school, Xavier, officially retired his uniform number 9 in a commemoration on January 30, 1999. As the "Killer B's" brand gained increased national attention, journalist Dayn Perry jocosely noted that in 1999 the Astros, "in pursuit of arcane history, used eight players whose last names began with 'B'", including Bagwell, Paul Bako, Glen Barker, Bell, Sean Bergman, Lance Berkman, Biggio, and Tim Bogar. On April 21, Bagwell hit three home runs in a 10–3 win against the Chicago Cubs at Wrigley Field, his second career three-home run game. The second home run allowed him to overtake Wynn as the Astros' all-time home run leader at 224 and he tied a career-high with six RBI in one game. Bagwell produced another three-home run game on June 9 against the Chicago White Sox that was a grand slam short of the "home run cycle", accounted with a solo home run, a three-run home run, and a two-run home run, respectively. The two three-home run games made him the only player to accomplish this feat at two different stadiums in Chicago in the same season. Nominated to his fourth career All-Star Game, Bagwell to that point in the season had scored or driven in 28.6% of the Astros' runs, the highest portion of a team's offense for which any one player in MLB accounted. He was first in the NL in walks (83), runs scored (81) and OBP (.464), second in home runs (28), RBI (78) and SLG (.648) and stole 17 bases while Houston remained percentage points behind Cincinnati for first in the division.
=== Remote influence on tropical Atlantic Ocean === A study of climate records has shown that El Niño events in the equatorial Pacific are generally associated with a warm tropical North Atlantic in the following spring and summer. About half of El Niño events persist sufficiently into the spring months for the Western Hemisphere Warm Pool to become unusually large in summer. Occasionally, El Niño's effect on the Atlantic Walker circulation over South America strengthens the easterly trade winds in the western equatorial Atlantic region. As a result, an unusual cooling may occur in the eastern equatorial Atlantic in spring and summer following El Niño peaks in winter. Cases of El Niño-type events in both oceans simultaneously have been linked to severe famines related to the extended failure of monsoon rains.

It binds and activates both the GIP and GLP-1 receptors, making it a dual incretin receptor agonist. Single-receptor GLP-1 agonists act on one target only. The dual profile is the defining pharmacological feature of the molecule.
A fatty diacid side chain promotes reversible binding to serum albumin. This association slows renal clearance and protects the peptide from rapid enzymatic degradation. The result is a prolonged circulation time that supports weekly administration.
The first regulatory approval, for type 2 diabetes, was granted in the United States in 2022. An additional approval for chronic weight management followed in 2023. Availability and approved indications vary by country and are set by each national regulator.
Reversed-phase high-performance liquid chromatography is the standard approach, separating the main peak from related impurities. Ultraviolet detection near 214 nanometers captures the peptide backbone. Mass spectrometry is then used alongside chromatography to confirm identity and detect covalent modifications.
